    Research: "Crystallization of Single Stranded Annealing Proteins in a DNA Complex"

    Research: Single-stranded annealing proteins (SSA) bind to single-stranded DNA (ssDNA) and are involved in pairing homologous DNA and double-stranded breaks. The RecT protein from Escherichia coli binds to ssDNA and is involved in recombination and in promoting the annealing of complimentary DNA strands. For all eukaryotes, Rad52, a 400 residue protein, promotes the same reactions as RecT and $\beta$, and is involved in DNA repair, synthesis and multiple DNA repair pathways. Cells and DNA have mechanisms to repair mutations; however, failure to do this may lead to cancer. Therefore, it is important to understand how the DNA repair processes of RecT and $\beta$ work so that such mistakes can be prevented. Although, through many studies, some functions of these proteins have been revealed, the structures of RecT and $\beta$β protein has not been solved. This study aims to determine experimentally the mechanism by which SSA proteins bind to ssDNA and promote the annealing of complimentary DNA. The purpose of this study is to study how proteins recognize and repair damaged sites of DNA in cells. We use x-ray crystallography and diffraction to solve the three-dimensional atomic structure of RecT and $\beta$ protein in complex with DNA substrates.

    Research: i) "Cervical Cancer Treatment research", Radiation Oncology Dept, OSU, Advisors: Dr. Nina Nayr and Dr. Huang
    "Study of cervial cancer using MRI": MRI was used to monitor tumor regression and predict the radiation therapy (RT) outcome. I collected data of tumor volume and dosimetry from MRI scans before, during, and after RT of 35 patients and recorded into Microsoft Excel for fast data analysis. A kinetic model was used to describe tumor response to RT. The model incorporated three major effects (radiation cell killing, dead-cell resolving, and tumor repopulation) and was used to analyze the tumor regression data. The model parameters derived for individual patients correlated with therapy outcome. New methods based on current findings can be used to predict clinical outcome and adaptive therapy can be adopted for patients at risk of treatment failure
