    RESEARCH EXPERIENCE

    6. OSU Clinical Lab, OSU Hospital Columbus, OH
Advisors: Professor Clay Marsh and Prof. Melissa Piper
    Dec. 2012 – Jan. 2013
  • studied expression of RNA (microRNA and mRNA) and the microvesicles from serum/plasma of patient blood samples and looked for biomarkers
  • used realtime PCR cloning and genotyping for a mouse hypoxia-inducible factor (HIF) study

    5. Cornell Protein Crystallography: Investigation of vicbrio cholera matrix proteins
Ithaca, NY, Cornell University, Department of Molecular Medicine
    Summer 2012 – present
    Advisor: Professor Holger Sondermann

  • studied Bap1 and RbmC, two extracellular matrix proteins in V. cholerae through crystallography by finding proper constructs for their expression and determining their crystal structure and protein functions
  • chose primers for protein expression, used PCR for cloning, and then performed ligations and transformation for expression
  • purified the proteins that were able to expressed through chromatography
  • the purified products are used in further steps of crystallography: screening using crystallization sets to get initial crystals, optimization, and x-ray diffraction at the Cornell High Energy Synchertron
  • solve structures by modeling with HKL2000 and Phenix using electron density map

    4. Cellulase and Protein Engineering Research
Ithaca, NY Cornell University, Department of Molecular Biology and Genetics
    Summer 2012
    Advisor: Professor David Wilson
    Research:

  • DNS assay experiments (1 and 24 hour) using Cel5A enzyme and BMCC cellulose o Some experiments used antibodies to study their effect in inhibiting enzyme activity
  • Created models of Cel5A using Pymol and MDGTools, by adding glucose cleaving ligand to active site of enzyme

  • Cornell Research, 2011-2012: Advisor: Prof. H. Sodermann

    3. Spring 2011 (for OSU research credits) - from March 2010:

Advisor: Professor C. Bell, Molecular and Cellular Biochemistry Dept
Research: "Crystallization of Single Stranded Annealing Proteins in a DNA Complex"

Research: Single-stranded annealing proteins (SSA) bind to single-stranded DNA (ssDNA) and are involved in pairing homologous DNA and double-stranded breaks. The RecT protein from Escherichia coli binds to ssDNA and is involved in recombination and in promoting the annealing of complimentary DNA strands. For all eukaryotes, Rad52, a 400 residue protein, promotes the same reactions as RecT and $\beta$, and is involved in DNA repair, synthesis and multiple DNA repair pathways. Cells and DNA have mechanisms to repair mutations; however, failure to do this may lead to cancer. Therefore, it is important to understand how the DNA repair processes of RecT and $\beta$ work so that such mistakes can be prevented. Although, through many studies, some functions of these proteins have been revealed, the structures of RecT and $\beta$β protein has not been solved. This study aims to determine experimentally the mechanism by which SSA proteins bind to ssDNA and promote the annealing of complimentary DNA. The purpose of this study is to study how proteins recognize and repair damaged sites of DNA in cells. We use x-ray crystallography and diffraction to solve the three-dimensional atomic structure of RecT and $\beta$ protein in complex with DNA substrates.

    2. Under NIH supported SREP at CCTS, Ohio State University, Summer 2009 (after 10th Grade):

Research: i) "Cervical Cancer Treatment research", Radiation Oncology Dept, OSU, Advisors: Dr. Nina Nayr and Dr. Huang
"Study of cervial cancer using MRI": MRI was used to monitor tumor regression and predict the radiation therapy (RT) outcome. I collected data of tumor volume and dosimetry from MRI scans before, during, and after RT of 35 patients and recorded into Microsoft Excel for fast data analysis. A kinetic model was used to describe tumor response to RT. The model incorporated three major effects (radiation cell killing, dead-cell resolving, and tumor repopulation) and was used to analyze the tumor regression data. The model parameters derived for individual patients correlated with therapy outcome. New methods based on current findings can be used to predict clinical outcome and adaptive therapy can be adopted for patients at risk of treatment failure

ii) Continued research on radiation dosemetry for uniform irradiation and wrote the draft paper; Advisor: Dr. J. Wang

    1. "New approach for reduction of hot and cold spots in craniospinal irradiation" {\bf Research Student}, Summer 2008 (after 9th Grade)-present, Radiation Oncology Dept, James Cancer Hospital, OSU, Advisors: Professor. Jian Wang, Dr. Montebello

Research: "Radiation Dosimetry". On finding novel approaches for reduction of hot and cold spots and uniform irradiation on fluids in craniospinal area for cancer treatment. The aim is to have more effective radiation therapy in contrast to the conventional method which causes hot and cold spots making the treatment less effective. The first phase (complete) - study through simulation experiments using the biomedical code ECLIPSE, next phase - implementation on patients (11th Grade)
